Dentin hypersensitivity (DH)
Dentin hypersensitivity (DH) is a sharp and short duration dental pain, arising from exposed dentin surfaces in response to different type of stimuli. Overall, DH is estimated to affect to some degree more than 20% of the general population. It is a feed-forward process characterized by the loss of the enamel and dentin due to dental demineralization by acid foods, cariogenic bacteria and the activation of dentine Matrix Metalloproteinases (MMPs) that follows dental demineralization. The opening of dentinal tubules allows the movement of internal fluids and results in pain.
Which technologies, which products?
PolymP®-Active (Zn) are Zinc functionalized polymer spherical particles with size ranging between 200 nm and 250 nm. From a regulatory standpoint, the product could be developed as an Oral Care Product. The particles have as main features:
- Highly bioactive and contribute to fast dentin remineralization by enhancing the precipitation of natural minerals that are not dissolved by saliva.
- Occlude dentin tubules in a very efficient way.
- Increase dentin mechanical properties, then dentin will become resistant to subsequent abrasion.
- Possess antibacterial activity.
- Inhibit dentin metalloproteinases, preventing the accelerated destruction of dentin.
- Low Mohs hardness: Do not result in dentin or enamel abrasion.
Regulatory studies (Acute Oral Toxicity, Gingival Epithelium in vitro Irritation Study, Skin Sensitization and Ames Test) carried out by a CRO under GLP have demonstrated that PolymP®-Active (Zn) has an excellent safety profile. Moreover, large batches can be produced under GMP conditions.
Applications & Market
Topical application of these particles may find application for the treatment of dentin hypersensitivity. Major expected advantages versus the main competitors are the irreversible filling of dentine tubules and the lack of abrasion.
These particles and their potential therapeutic applications are protected by a recent PCT patent application. Taking into account the prevalence of dental hypersensitivity we consider that there are very large market opportunities for our agent. In 2015 the sensitivity toothpaste market was worth €900 million.
